Bring an electric converter if you use a hair dryer, electric shaver, phone charger, etc (can get one at any electronic store like Radio Shack). Definitely bring comfortable walking shoes (may help to bring a pair that is comfortable for walking long distances and can double as a semi-nice shoe like for dinner out - helps to keep you from over packing!). Make sure to watch the weather forecasts before you go and pack accordingly. I don't like the bath towels in hotels in Europe (hard and itchy) so I bring an old one from home and just leave it there when I leave. Remember you can even hand wash some clothing items in the sink at a hotel (most hotels have laundry services for a fee), so try not to over-pack! Go to a local travel agent and pick up some brochures of the places you are going so you can get an idea of want you want to see while you are there. I'm sure you will have a wonderful time - sounds like a great trip!
Pack as light as possible, and items that have double uses,or that you can layer, a lightweight raincoat that you can carry easily in a bag that you would use for the day, just in case you get caught in a shower.
Try not to go for fabrics that crease a lot.
Comfortable shoes are a must.
Depending on the amount of time you are traveling for and how much time you have, I tend to carry the minimum of toiletries as these are heavy. If you are moving from hotel to hotel, most of these provide shampoo etc, so you only need to take sample sizes with you. If your backpacking then think about the amount you need for the time, they sell toiletries overseas and you can ditch them when you leave.
Lots of memory cards or film for cameras, it may be worth getting a universal battery charger as well.
In Barcelona, Istanbul, be careful how you carry your bag when walking around as there is a lot of snatch and grab. So it may be worth getting a money belt that you can wear under your clothes.
